本试剂盒采用双抗体夹心酶联免疫吸附法定量检测小鼠IL-1β Pro-form。小鼠IL-1β Pro-form试剂盒仅供科学研究,不用于临床诊断。白细胞介素 1 beta (IL-1β)是由单核细胞、巨噬细胞和树突状细胞表达的促炎性细胞因子。 机体对炎性刺激物产生应答后,合成分子量为 31 kDa 的无活性的 IL-1β 前体,累积于细胞质中。 裂解 pro-IL-1β 为有活性的分子量为 17 kDa 的蛋白需要炎性体的激活。炎性体的激活触发 caspase-1前体加工成它的活性形式, 反过来裂解 pro-IL-1β。虽然 IL-1β 最常以活性形式被分泌存在, 但是未裂解蛋白的分泌在一些生物条件下也可被检测到。IL-1β 通过两个受体 IL-1RI 和 IL-1RII 进行信号传导。这些细胞因子通过触发靶细胞中其它促炎性细胞因子的产生和启动急性期应答, 在宿主的先天免疫防御中发挥重要作用。它们的活性受IL-1 受体拮抗剂(IL-1RA)调控,后者是一个由多细胞类型产生的、 可通过竞争性抑制来阻断受体结合的蛋白。 IL-1β 水平升高与许多慢性炎症状态相关, 这意味着 IL-RA 或者 IL-1β 中和抗体有潜在的治疗价值。

IL1B 分子靶点综述
白细胞介素1(IL-1)是17kDa的胞外多肽,分为IL-1α和IL-1β两种蛋白。IL-1β是活化的巨噬细胞分泌的蛋白前体,通过caspase1(CASP1/ICE)水解产生的活化形式。该细胞因子是炎症反应的重要介导者,参与许多细胞活性,包括细胞增殖、分化和凋亡。IL-1是一类重要的蛋白家族,它们由生物活化的单核细胞所衍生,参与炎症反应和免疫应答。IL-1β表达升高会产生各种不同的自身炎症综合征,最显著的是单基因条件称为CAPS,是由于触发IL-1β加工的炎性体受体NLRP3发生突变而引起的。本试剂盒用来检测人IL-1β的成熟形式和未裂解的前体形式。
