| 背景介绍 | 白细胞介素6(IL-6)是一种同时具备促炎性和抗炎性的细胞因子。成年小鼠IL-6由187个氨基酸组成,与人类和大鼠白IL-6共有39%和85%的序列同源性。
在感染期间和受到外伤后,尤其是烧伤或其它组织损伤导致的炎症,IL-6会由T细胞和巨噬细胞分泌,促进免疫应答。研究表明,小鼠抵抗肺炎链球菌感染时需要IL-6的参与,说明IL-6在抵抗感染中发挥作用。 IL-6在很多疾病如糖尿病、动脉粥样硬化、抑郁症、阿尔茨海默氏病、系统性红斑狼疮、多发性骨髓瘤、前列腺癌、白塞氏病和类风湿性关节炎等中可促进炎性和自身免疫疾病进程。晚期/转移性癌症患者的血液中具有较高水平的IL-6。 |
| 检测原理 | 本试剂盒采用双抗体夹心酶联免疫吸附检测技术。特异性抗小鼠IL-6抗体预包被在高亲和力的酶标板上。酶标板孔中加入标准品、待测样本和生物素化的检测抗体,经过孵育,样本中存在的IL-6与固相抗体和检测抗体结合。洗涤去除未结合的物质后,加入辣根过氧化物酶标记的链霉亲和素(Streptavidin-HRP)。洗涤后,加入显色底物TMB,避光显色。颜色反应的深浅与样本中IL-6的浓度成正比。加入终止液终止反应,在450 nm波长(参考波长570 - 630 nm)测定吸光度值。 |
