本酶在以ATP作辅酶的情况下，可以催化相邻DNA链的5’磷酸基团和3’羟基之间的连接反应。平末端和粘性末端均可，此酶也可以催化双链RNA与双链DNA连接，但不能催化单链核酸的连接。
储存条件：
T4 DNA连接酶和10×T4 DNA ligase Buffer必须放置于-20℃保存，避免反复冻融，10×T4 DNA ligase Buffer建议分装使用。
