使用说明: 1. 对于固定的细胞或组织: (1) 贴壁细胞 a. 取普通洁净盖玻片于70%乙醇中浸泡5min或更长时间,用细胞培养基PBS或0.9%NaCl洗涤3遍,再用细胞培养液洗涤1遍。将盖玻片置于6孔板内,种入细胞培养过夜,使约为50%-80%满。 b. 刺激细胞发生凋亡后,吸尽培养液,加入0.5ml固定液,固定10min或4℃过夜。 c. 去固定液,用 PBS或0.9%NaCl洗涤2遍,每次3min,吸尽液体。 d. 加入0.5mlHoechst 33258染色液,染色5min。也宜用摇床,或手动晃动数次。 e. 用PBS或0.9%NaCl洗涤2遍,每次3min,之后用抗淬灭封片液封片。 (2)悬浮细胞 a. 离心收集细胞样本于1.5ml离心管内,加入0.5ml固定液,缓缓悬起细胞,固定10min或4℃过夜。 b. 离心去固定液,用PBS或0.9%NaCl洗涤2遍,每次3min。 c. 离心后吸去大部分液体保留约50μl液体,再缓缓悬起细胞,滴加至载玻片上,尽量使细胞分布均匀。 d. 稍晾干,使细胞贴在载玻片上不易随液体流动,均匀滴上0.5mlHoechst 33258染色液,染色5min。用吸水纸从边缘吸去液体,微 晾干。 e. 用PBS或0.9%NaCl洗涤2遍,每次3min,之后用抗淬灭封片液封片。 (3)组织切片 对于任何常见切片,处理至常规可以进行免疫染色时,或完成常规的免疫染色后,用PBS或0.9%NaCl洗涤2遍,每次3min,即可进行后续的Hoechst 33258染色,染色5-10min,用PBS或0.9%NaCl洗涤2遍,每次3min,之后用抗淬灭封片液封片。 2. 对于活细胞或组织: a. 加入适当量Hoechst 33258染色液,必须充分覆盖住待染色的样品,通常对于六孔板一个孔需加入1ml染色液,对于96孔板一个孔 需加入100μl染色液。 b. 在适宜于细胞培养的温度培养20-30min。弃染色液,用PBS或培养液洗涤2-3次即可进行荧光检测。
