Description 
Peroxidase-antiperoxidase complex consisting of horseradish peroxidase and immunospecifically pure rat anti-peroxidase-Rat clonoPAP® Clone SMI 4070 Form Purified Antibody (in PBS, 0.01M sodium azide) Host Rat Specificity ACTIVITY-SELECT Rat ClonoPAPRat ClonoPAP® is intended for the detection of monoclonal or serum-derived rat antibodies via an excess of an anti-rat IgG (such as goat antirat IgG Cat # SMI 5040). Rat antibodies of any isotype are detectable via reaction with shared light chains.
Composition: Immune complex of horseradish peroxidase and monoclonal rat antiperoxidase providing maximal immunocytochemical staining intensity at a dilution range of 1:100 to 1:6,400. Staining intensity decreases at dilutions of 1:12,800 and beyond.Uses This antibody is primarily used for Immunohistochemistry applications, but it is also effective in blotting and ELISA assays. It can be used with diaminobenzidine tetrahydrochloride (DAB) substrates for detection with peroxidase. Suggested Working Dilution ACTIVITY-SELECT Rat ClonoPAP Recommended dilutions for rat ClonoPAP® range from 1:100 to 1:6,400 in its use in immunocytochemistry and other immunodetection systems. Extend of optimal dilution depends upon nature and concentration of the antigen examined, species of the antigen, mode of fixation, kind of section used in immunocytochemistry, as well as the activity of the second antibody (anti-IgG) employed. In many cases increasing dilutions of rat ClonoPAP® will not decrease staining intensity, and may occasionally even increase it, until a point of excessive dilution is reached. The optimal dilution is the highest or next-to-highest dilution that still yields maximal staining. Dilutions of ClonoPAP® more concentrated than 1:100 are not recommended. Concentrations higher than those recommended may produce a brown background and decrease specific staining intensity. Dilutions should be carried out in buffer containing 1% normal goat serum when rat ClonoPAP® is used in conjunction with goat antirat IgG. Notes This antibody is sold for research use only, not for human or in-vivo use. Covance antibodies may not be resold or modified for resale without prior written approval. Storage Store at -20°C. Upon initial thawing, apportion into working aliquots and store at -20°C. Avoid repeated freeze-thaw cycles to prevent denaturing the antibody.
