Description: Echelons Sphingosine Kinase 1 Activity Assay is an ATP depletion assay which quantitates the remaining ATP levels in solution following the kinase reaction. The assay reaction is prepared by the addition of sphingosine kinase, sphingosine (substrate) in reaction buffer and initiated by the addition of ATP. The reaction is then stopped by adding the ATP detector after the chosen reaction time. The luminescent signal is inversely correlated with the kinase activity. Echelons sphingosine kinase 1 activity assay is tested with Echelons SPHK1 (E-K068) and also worked with SPHK2 (E-K069)
Sphingosine kinases (SPHKs) catalyze the phosphorylation of sphingosine to sphingosine-1-phosphate (S1P). SPHKs have been implicated in proliferation, survival, migration and regulation of Ca2+homoeostasis, development and regulation of the cardiovascular and nervous systems, inflammation, immunity, and cancer growth. There are two currently known SPHK isoforms, SPHK1 and SPHK2. It is believed that SPHK1 and SPHK2 have opposite functions, promoting cell growth and apoptosis respectively.
