Cytotoxicity of natural killer cells is regulated by a balance of signals from two kinds of NK receptors, activating receptors and inhibitory receptors. Natural cytotoxicity receptors (NCR) family, which consists of NKp30, is a major human activating NK receptor. NKp30, also known as NCR3 and CD337, is the third identified 30-kD triggering receptor belonging to a major human activating NK receptor family, termed as natural cytotoxicity receptors (NCR), which also contains NKp44 and NKp46. As a type I transmembrane protein having a single extracellular V-like immunoglobulin domain, NCR3 is selectively expressed by all resting and activated human NK cells, and plays a key role in NK-mediated killing of most tumor cells. In addition, NKp30 is also involved in NK-mediated induction of dendritic cell (DC) maturation. NKp30 forms a complex with an ITAM-bearing accessory protein CD3-zeta which undergoes phosphorylation upon a specific antibody ligation. It has been demonstrated that NK cell activation signaling specifically induces transcriptional activation and synthesis of new NF-κB dependent proteins during the initiation of cytotoxicity.
