应用:
胶原酶已用于研究晚期糖基化终产物(AGE)所需的动脉组织的制备。该酶还与其他蛋白酶一起用于人肿瘤、小鼠肾、人脑、肺上皮以及许多其他组织的解聚。它还可有效用于肝脏和肾脏的灌注研究、胰腺的消化以及非实质肝细胞的分离。该酶经过了肝细胞释放的适用性测试，针对每只大鼠肝脏的100ml总体积中的浓度大约为1mg/mL。
来自组织溶梭菌的胶原酶已被用于:
●制备用于消化肿瘤组织的胶原酶溶液；
●作为磷酸盐缓冲生理盐水的补充剂用于消化已切除的肠系膜脂肪组织；
●作为罗斯韦尔公园纪念研究所1640培养基(RPMI-1640)的补充剂用于消化结肠段；
●用于消化子宫内膜组织；
●制备用于消化去被膜的睾丸的酶A和酶B。
生化/生理作用:组织溶梭菌产生两类胶原酶。梭菌胶原酶比哺乳动物胶原酶更高效。它在胶原三螺旋中的多个裂解位点进行裂解。梭菌胶原酶与气性坏疽中的细菌入侵有关。从组织中进行细胞的有效释放需要胶原酶和中性蛋白酶的作用。每摩尔的胶原酶可被四克的钙原子(Ca²⁺)所激活。培养滤液中被认为至少含有7种分子量在68-130kDa之间的不同蛋白酶。最适pH为6.3-6.8。该酶通常用于消化组织样品中的结缔成分以释放单个细胞。胶原酶处理可引起某些细胞的死亡。通常，使用0.1至5mg/ml的浓度用于消化。反应时间可从15分钟至数小时，并在不引起过多细胞死亡的情况下达到令人满意的细胞解离效果。优选含有钙和BSA的Krebs Ringer缓冲液，而且Zn²⁺是活性所必需的。每摩尔胶原蛋白酶被4g原子钙活化。它被乙二醇-双(β-氨基乙基醚)-N，NN′,N′-四乙酸，β-巯基Ji乙醇、谷胱甘肽、巯基Ji乙酸和8-羟基喹Kui啉抑制。
注意:以原包装形式，该产品在-20°C下可稳定保存6年。在37℃、50℃和-20℃下30天内，FALGPA或蛋白酶活性没有损失。如果分液(10mg/ml)形式快速冷冻并且在-20℃下冷冻保存，粗胶原酶的溶液可保持稳定。进一步的冻融步骤会破坏溶液稳定性。当在冰上保持时，该产品可在7小时内保持100％活性。
