一般描述:分散酶是一种中性、快速反应、温和的蛋白酶，能将表皮从真皮中完整分离。它还能将培养的上皮细胞从基质中完整分离出来。其可分裂基底膜区，使上皮细胞保持完整。
特异性:分散酶是一种非特异性的金属蛋白酶。
应用:分散酶®用于从多种不同的组织和器官制备细胞。该酶已经证实是一种快速、有效但温和破坏组织胞外基质的制剂，可释放用于细胞培养的单细胞(原代细胞培养)或收集培养细胞以转至新底物中(继代培养)。此外，它还可用来防止细胞悬浮培养过程中形成不必要的团块。

单位定义:一单位是指在pH 7.5、+37°C反应条件下，在1分钟内释放出对应1μmol(181μg)酪氨酸的Folin-阳性的氨基酸和肽类所需的酶量。
Roche分散酶®II单位与日本文献中引用的分散酶单位(其中分散酶®浓度为1000到2000单位/ml的情况不常见)的实际比较结果表明，Roche的1单位分散酶大致相当于日本的600单位。
Roche Applied Science的1单位®相当于181蛋白酶单位(PU)，其中PU为在pH 7.5和+37°C条件下，在1分钟内释放相当于1μg酪氨酸的氨基酸所需的酶量。
制备说明:
激活剂:最佳Ca2+浓度为2mM。酶制剂含有维持最佳活性所需的足量Ca2+。
抑制剂:EDTA、EGTA、Hg2+、其他重金属。分散酶不受血清抑制。
工作浓度:0.6至2.4U/ml
工作溶液:储备溶液和工作溶液制备:
制备10mg/ml的储备溶液，将分散酶®II冻干粉溶于HEPES-缓冲盐溶液中(50mM HEPES/KOH pH 7.4，150mM NaCl)。制备工作溶液，将上述储备溶液用分离细胞培养基稀释至终浓度为0.6到2.4U/ml。请注意，建议浓度不要高于2.4U/ml。为实现最佳结果，用0.22μm滤膜过滤工作溶液。
储存条件(工作溶液):-15至-25°C
复溶的储备溶液在2至8℃下可稳定保存2周。如需保存2个月以上，储备溶液需分装冻存。避免反复冻融! 采用PBS稀释的工作溶液在2至8℃下可稳定保存3天。
储存及稳定性:2至8°C储存。(干燥储存!)
