BCA蛋白浓度测定试剂盒(500T)产品简介：Bicinchoninic acid (BCA )法是在世界上常用的蛋白浓 度检验方法之一BCA法基础上改进而成。其原理与Lowery法蛋 白定量相似，即在碱性环境下蛋白质与Cu2+络合并将Cu2+还原成 Cu+。两分子BCA与一个Cu+螯合形成稳定的紫蓝色复合物，在 562 nm处有高的光吸收值并与蛋白质浓度成正比，据此可测定蛋 白质浓度。与Lowery法相比，BCA蛋白测定方法灵敏度高，操作 简单，试剂及其形成的颜色复合物稳定性俱佳，在50-200μg/ml 浓度范围内有良好的线性关系，并且受干扰物质影响小。与 Bradford法相比，BCA法的显著优点是不受去垢剂的影响。

产品简介：
碱性条件下，蛋白将Cu 2+ 还原为Cu + ， Cu + 与BCA试剂形成紫蓝色的络合物，测定其在 562nm处的吸
收值，并与标准曲线对比，即可计算待测蛋白的浓度。常用浓度的去垢剂SDS，Triton X-100，Tween不影响
检测结果，但受螯合剂(EDTA，EGTA)、还原剂（DTT，巯基乙醇）和脂类的影响。实验中，若发现样品稀
释液或裂解液本身背景值较高，可试用Bradford蛋白浓度测定试剂盒。
操作说明：
一 . 微孔酶标仪法
1. 配制工作液: 根据标准品和样品数量，按 50 体积 BCA 试剂加 1 体积 Cu 试剂（50:1）配制成 BCA
工作液，充分混匀(混合时可能会有浑浊，但混匀后就会消失)。BCA 工作液室温 24 小时内稳定。
2. 稀释标准品：取 10 微升 BSA 标准品用 PBS 稀释至 100 微升（样品一般可用 PBS 稀释） ，使终浓度
为 0.5mg/ml。将标准品按 0, 2, 4，6，8, 12, 16, 20 微升加到 96 孔板的蛋白标准品孔中，加 PBS 补足到 20
微升。
3. 将样品作适当稀释（最好多做几个梯度，如作 2 倍、4 倍、8 倍稀释） ，加 20 微升到 96 孔板的样
品孔中。由于移液器在取小量时的误差，标准线前面的点可能不很准确，所以尽可能的让样品点落在标准
线 1/2 后。
4. 各孔加入 200 微升 BCA 工作液，37℃放置 15-30 分钟。用酶标仪测定 A562nm，根据标准曲线计
算出蛋白浓度。使用温箱孵育时，应注意防止因水分蒸发影响检测结果。
二 . 分光光度计法
如没有酶标仪，可用分光光度计在离心管中混匀后加入比色皿中比色。
步骤如下：
1. 配制工作液: 根据标准品和样品数量，按 50 体积 BCA 试剂加 1 体积 Cu 试剂（50:1）配制成 BCA
工作液，充分混匀(混合时可能会有浑浊，但混匀后就会消失)。BCA 工作液室温 24 小时内稳定。
2. 稀释标准品：取 100 微升 BSA 标准品用 PBS 稀释至 1ml（样品一般可用 PBS 稀释） ，使终浓度为
0.5mg/ml。
3. 取八支（或者更多）5ml 离心管，标上号，按下表加入试剂。

4. 37℃放置 15-30 分钟。用分光光度计测 562nm 处吸光值，根据标准曲线计算出蛋白浓度。
注意事项：
