| 描述 | Telithromycin is a semisynthetic 14-membered macrolide antibiotic and erythromycin derivative that binds to the 50S subunit of the 70S bacterial ribosome, inhibits bacterial protein synthesis, and exhibits antimicrobial activity against a wide range of gram-positive and gram-negative bacteria for the treatment of mild to moderate respiratory infections. |
| 体外活性 | 采用微量肉汤稀释法、琼脂稀释法、E-test法等方法测定了Telithromycin的体外抗菌活性。其MIC分布均匀（MIC范围为0.003～0.25 μg/ml）[1]。但其对鸟分枝杆菌复合体的MIC50和MIC90均大于16 μg/ml，表明其体外抗菌活性较弱[2]。 |
| 体内活性 | 小鼠模型显示，口服Telithromycin（3-26mg/kg）对耐红霉素A败血症具有卓越的疗效，并对多种致病菌表现出显著的抗菌作用。在耐红霉素A肺炎链球菌模型中，Telithromycin的ED50为6.5mg/kg，其疗效与阿奇霉素相当。在100mg/kg剂量下，Telithromycin的血浆Cmax达到8.5mg/L，肺浓度为13.3μg/g，分布体积接近人体，血浆和肺组织的半衰期分别为2.5小时和2.8小时[3]。 |
