| 名称 | Bicuculline |
| 描述 | Bicuculline is an alkaloid extracted from Corydalis decumbens, acting as a competitive antagonist of the neurotransmitter GABAA receptor (IC50 = 2 μM). Bicuculline also blocks Ca2+-activated potassium (SK) channels and inhibits slow afterhyperpolarization (slow AHP). Bicuculline has anticonvulsant effects and is commonly used to establish mouse seizure models. |
| 体外活性 | 方法：小鼠海马脑片 CA1 锥体神经元 （WT vs PGC-1α−/−），通过浴槽给药方式加入 Bicuculline （1 μM），稳定灌流后全细胞电压钳记录计算 I/E 比率。 结果：PGC-1α−/− 双突触 IPSCs 幅度约为 WT 的 2 倍，I/E 比率显著升高，而 Bicuculline 将 PGC-1α−/−的 IPSCs 和 I/E 比率恢复至 WT 水平，且不影响 EPSCs。[1] 方法：新生小鼠（P5）小脑培养星形胶质细胞加入 Bicuculline （100 μM）和 GABA （50 μM），GABA 应用 30 s，间隔 7 min，全细胞膜片钳记录电流。 结果：荷包牡丹碱（100 μM） 能阻断（90.5 ± 3.7%）50 μM GABA 诱发的电流。[2] |
| 体内活性 | 方法：雄性 SD 大鼠通过皮下注射 Bicuculline (1， 4 mg/kg)，给药后 10， 30， 110 min 取血和脑组织，LC-MS/MS 测定血浆和脑组织药物浓度；观察记录惊厥等级。 结果：Bicuculline 呈剂量依赖性进入脑内，1 mg/kg 组脑内 Tmax 为 10 min，4 mg/kg 组为 30 min，脑内浓度>880 ng/g 时诱发惊厥(≥III 级)。[2] |
