| 描述 | Piritrexim (BW 301U) is an orally available fat-soluble dihydrofolate reductase inhibitor with pulmonary toxicity used in the study of uroepithelial carcinoma and metastatic breast cancer. |
| 体外活性 | Piritrexim（0.1至1.0 microM）能够在小鼠腹膜巨噬细胞模型中抑制T.gondii的复制。单独使用时无效的sulfadiazine与piritrexim联用，可在更低的piritrexim浓度下抑制T.gondii复制。[5] |
| 体内活性 | Piritrexim (25-mg/m2/剂量；口服) 在4位患者中引起了骨髓抑制和黏膜炎，但在接受15和20-mg/m2/剂量治疗的患者中并未见此反应。Piritrexim (20 mg/m2/剂量；口服) 吸收迅速，口服后平均达到峰值浓度的时间为1.5小时，而浓度-时间曲线下面积（AUC）与给药剂量线性相关。槽谷血浆Piritrexim浓度与DLT（剂量限制毒性）强烈相关（P = 0.0016）。槽谷血浆Piritrexim浓度大于0.5 microM似乎能预测毒性。超过此阈值的槽谷浓度的15位患者中有11位经历了DLT。 |
