| 描述 | Rupintrivir (AG7088) is an antiviral agent and an irreversible inhibitor targeting human rhinovirus 3C protease (EC50 = 0.023 μM), featuring high selectivity and immunomodulatory activity, capable of effectively inhibiting multiple HRV serotypes as well as EV71 and EV-D68 replication, commonly used for research on viral infection mechanisms. |
| 体外活性 | 方法：将 RD 细胞与梯度浓度 Rupintrivir 孵育 3–4 天。 结果：Rupintrivir对 EV-D68、RV-87 及 EV-71 均有强效抑制，其中对 EV-D68 的 EC₅₀ 为 0.0022–0.0053 μM，EC₉₀ 为 0.004 μM，为十种化合物中活性最强。[1] 方法：采用荧光共振能量转移法（FRET assay），将梯度浓度 Rupintrivir 与纯化的 DHAV 3C 蛋白酶及荧光底物共孵育 2 小时。 结果：其对鸭甲肝病毒 3C 蛋白酶具有抑制活性。[2] |
| 体内活性 | 方法：采用HDM致敏Balb/c小鼠制备精密肺切片，离体感染RV1b（10⁵ TCID₅₀/mL），并以100 nM Rupintrivir处理48 h。 结果：Rupintrivir显著抑制HDM致敏组中RV诱导的IL-4和IL-6过度分泌，对干扰素应答无影响。[3] |
