| 描述 | Candoxatril (UK 79300) is a small-molecule inhibitor and a neutral endopeptidase (NEP) inhibitor with oral activity and cell permeability, used for the treatment of chronic heart failure and capable of improving exercise capacity in patients receiving angiotensin-converting enzyme inhibitor therapy. |
| 体内活性 | 方法：采用单侧输尿管梗阻（UUO）诱导的肾纤维化小鼠模型，每日口服Candoxatril（剂量未详），连续给药1周。 结果：与溶剂对照组相比，Candoxatril显著增加尿cGMP水平，减少肾皮质胶原沉积（1.8%降至0.8%）和α-SMA表达（8.3%降至4.9%），表现出抗纤维化作用。[1] 方法：采用麻醉小鼠模型,经静脉注射给予 Candoxatril（5 mg/kg，溶于 0.9% NaCl），2 分钟后注射 GLP-1(9-36)NH₂。 结果：与溶剂对照组相比，Candoxatril 预处理组免疫反应性 GLP-1 浓度显著升高，表明其有效抑制 NEP 24.11 介导的 GLP-1 代谢物降解。[2] |
