| 描述 | Cefdinir (FK-482) is a semi-synthetic, broad-spectrum antibiotic in the third generation of the cephalosporin class, proven effective for common bacterial infections of the ear, sinus, throat and skin. |
| 激酶实验 | Topoisomerase II activity assay: Nuclear extracts are prepared, and nuclei are isolated. The activity of topoisomerase II is calculated from the percentage of decatenation obtained. Tritiated kinoplast DNA (KDNA 0.22 μg) is used as a substrate. Etoposide and topoisomerase II are incubated for 30 min at 37 ℃ and are stopped with 1% sodium dodecyl sulfate (SDS) and proteinase K (100 μg/mL). The percentages of decatenation and inhibition of topoisomerase II by Etoposide are obtained. |
| 体外活性 | Cefdinir 是一种新型口服 2-氨基-5-噻唑基头孢菌素，它能抑制人中性粒细胞在 PMA 刺激下的发光酚扩增化学发光（LACL）反应，但不能抑制氨溶酶原的反应。Cefdinir 抑制方式为浓度依赖性而非时间依赖性。Cefdinir 能抑制由水2、NaI和辣根过氧化物酶或含有氨酰过氧化酶的中性粒细胞提取物组成的无细胞系统中LACL的生成。Cefdinir 会损害钙离子拮抗剂 A23187 和 FMLP 诱导的 LACL 反应，这种损害在细胞松弛素 B 处理的中性粒细胞中会加剧。Cefdinir 直接抑制中性粒细胞受可溶性介质刺激时释放到细胞外介质中的含髓过氧化物酶的中性粒细胞提取物的活性，但对吞噬过程中释放到吞噬溶酶体中的提取物没有影响。[1] Cefdinir 对多种革兰氏阳性和革兰氏阴性细菌具有卓越的活性。Cefdinir 对多种 β-内酰胺酶具有耐药性，其 β-内酰胺稳定性通常优于头孢克洛和头孢呋辛。Cefdinir 主要通过肾脏排出体外。Cefdinir 与二肽转运体 PEPT1 和 PEPT2 相互作用。Cefdinir 肾小管重吸收量很大，Cefdinir 肾小管分泌可被丙磺舒抑制，这种分泌可能是由肾脏有机阴离子分泌途径介导的。[2] |
