实验流程概述（双抗体夹心法）

1. 把捕获抗体预包被的酶标板在室温平衡 30 分钟后使用。
2. 加入样本或标准品（建议双孔或三孔复孔），37 ℃孵育 1 h。
3. 洗涤（使用随盒提供的 Wash Buffer），随后加入生物素化检测抗体，37 ℃孵育 30 min。
4. 加入 Streptavidin‑HRP，继续 30 min 反应。
5. 再次洗涤后加入 TMB 底物（Substrate A + B），在暗处显色 10‑15 min。
6. 加入 Stop Solution，颜色转为黄；使用酶标仪在 450 nm（参考 620 nm）读取 OD 值。
7. 依据标准曲线计算样本中 IFN‑α 含量。

Assay Procedure Overview (Sandwich Immunoassay)

1. Allow the capture‑antibody‑coated microplate to equilibrate at room temperature for 30 minutes before use.
2. Add samples or standards (duplicate or triplicate wells recommended) and incubate at 37 °C for 1 hour.
3. Wash (using the Wash Buffer supplied) and then add biotinylated detection antibody; incubate at 37 °C for 30 minutes.
4. Add Streptavidin‑HRP and incubate for an additional 30 minutes.
5. Wash again, add TMB substrate (Substrate A + B), and develop in the dark for 10‑15 minutes.
6. Add Stop Solution; the color changes to yellow. Read absorbance at 450 nm (reference 620 nm) using a microplate reader.
7. Calculate IFN‑α concentrations in samples based on the standard curve.
