背景介绍

趋化因子配体11(CCL11)也称为嗜酸性粒细胞趋化因子(Eotaxin)，是CC趋化因子家族中的小分子量细胞因子。该因子通过诱导嗜酸性粒细胞的趋化性进行选择性招募，在反应过程中发挥重要作用。

研究表明，Eotaxin在人和小鼠血浆中的表达升高与衰老过程相关。在目前精神分裂症患者体内观察到更高浓度的Eotaxin表达水平。

检测原理

本试剂盒采用双抗体夹心ELISA法：

1. 特异性抗体预包被于酶标板
2. 样品或标准品中的CXCL11与固相抗体结合
3. 生物素化的检测抗体与结合的CXCL11特异性结合
4. 链霉亲和素-HRP偶联物与生物素化抗体结合

Background

Chemokine (C-C motif) ligand 11 (CCL11), also known as Eotaxin, is a small molecular weight cytokine belonging to the CC chemokine family. It plays a significant role in allergic processes by selectively recruiting eosinophils through the induction of eosinophil chemotaxis.

Studies indicate that elevated expression of Eotaxin in human and mouse plasma is associated with the aging process. Higher concentrations of Eotaxin have been observed in current cannabis users and patients with schizophrenia.

Assay Principle

This kit employs a sandwich ELISA principle:

1. A specific antibody is pre-coated onto the microplate.
2. CCL11 present in samples or standards binds to the immobilized antibody.
3. A biotinylated detection antibody specifically binds to the captured CCL11.
4. Streptavidin-HRP conjugate binds to the biotinylated antibody.
5. TMB substrate develops color catalyzed by HRP; the optical density (OD) is proportional to the CCL11 concentration.

Precautions

• Do not mix components from different kit lots.
• Equilibrate all components to room temperature for 30 minutes before use.
• Use disposable pipette tips to avoid cross-contamination.
