检测原理（双抗体夹心法）

1. 酶标板预包被特异性抗人vWF捕获抗体。
2. 加入待测样本或标准品，vWF 与固相抗体形成捕获复合物。
3. 洗涤除去未结合成分。
4. 加入HRP‑标记的检测抗体，形成双抗体夹心结构。
5. 再次洗涤后加入TMB底物，HRP 催化产生蓝色信号。
6. 加入终止液，颜色由蓝转黄，在 450 nm 读取 OD 值，峰值与浓度呈正比。

Assay Principle (Sandwich ELISA)

1. The microplate is pre‑coated with a specific anti‑human vWF capture antibody.
2. Test samples or standards are added; vWF binds to the solid‑phase antibody forming a capture complex.
3. Washing removes unbound material.
4. HRP‑conjugated detection antibody is added, creating a sandwich complex.
5. After a second wash, TMB substrate is added; HRP catalyzes a blue color development.
6. Stop solution is added, turning the color from blue to yellow. The absorbance at 450 nm is read; the signal is directly proportional to concentration.
