【产品概述】TA-克隆试剂盒适用于克隆带有 3’末端 A 突出的的 PCR 产物。试剂盒提供的pUCm-T 载体是为简化 PCR 产物的克隆而设计的，许多热稳定的 DNA 聚合酶，如 TaqDNA 聚合酶、Tth DNA 聚合酶等扩增的 PCR 产物在 3’末端后都带有一个突出的碱基 A，这样的 PCR 产物很容易连接到带有突出碱基 T 的 pUCm-T 载体上。 该载体的多克隆位点大多是单切点，并且调整了β-半乳糖苷酶读码框，更有利于克隆的蓝/白筛选。插入位点两端独特设计的两个 PstI 位点使插入片段可以用 Pst I 单酶切进行检测，也可以用非常廉价而高效的 EcoR I 和 Hind III 双酶切进行检测。可以用 M13 通用引物和 T7 启动子引物对 PCR 产物进行测序。pUCm-T 含有 T7 RNA 聚合酶的启动子，可以对插入片段进行体外转录。
