Ligation Mighty Mix在冰上融解，轻轻混合后再使用。 2. 长片段PCR产物（2 kb以上）进行DNA克隆时，连接反应时间请延长至数小时。 3. 克隆时，根据插入片段的长度和方向，有时即使插入了DNA片段也会有不出现终止密码或读码框不改变的情况发生，在蓝白选择培养基上会形成淡蓝色菌落。出现这种现象时，建议进行菌落PCR确认插入片段。 4. 进行切胶回收纯化DNA片段时要避免UV照射时间过长。长时间照射，DNA会受到损伤，影响克隆效率。 5. PCR用质粒模板与克隆载体 (pMD20-T vector；具有Ampr) 有同样的选择标记基因时，为防止出现质粒模板自身的转化菌落，将PCR反应液凝胶电泳后回收目的片段，纯化后再进行克隆。 | ||||||||||
