eIF2α, encoded by the EIF2S1 gene in humans, is the alpha subunit of the eukaryotic translation initiation factor 2 (eIF2) complex. This complex plays a pivotal role in the initiation of protein synthesis by facilitating the binding of the initiator tRNA (Met-tRNAᵢ) to the 40S ribosomal subunit, forming the ternary complex essential for translation initiation. eIF2 is a heterotrimer composed of three subunits: α (eIF2S1), β (eIF2S2), and γ (eIF2S3). A key regulatory aspect of eIF2α is its phosphorylation at serine 51. This phosphorylation is a critical component of the integrated stress response (ISR), a cellular mechanism that adjusts protein synthesis in response to various stressors such as amino acid deprivation, viral infection, or endoplasmic reticulum stress. Phosphorylation of eIF2α leads to a reduction in global protein synthesis, allowing cells to conserve resources and prioritize the translation of stress-responsive genes.
