ErbB3, also known as HER3 (Human Epidermal growth factor Receptor 3), is a member of the ErbB family of receptor tyrosine kinases, which includes EGFR (ErbB1/HER1), HER2 (ErbB2), and ErbB4 (HER4). Unlike other family members, ErbB3 possesses an impaired tyrosine kinase domain, meaning it has little to no intrinsic kinase activity on its own. However, it plays a critical signaling role by forming heterodimers, particularly with HER2, to activate downstream pathways. ErbB3 is a key player in cell growth, differentiation, survival, and migration, largely through the PI3K/Akt and MAPK/ERK pathways. It binds ligands such as neuregulins (NRG1/2) and, upon dimerization with kinase-active partners, becomes phosphorylated and transduces potent survival and proliferation signals. Due to its involvement in oncogenic signaling, drug resistance, and cancer progression—especially in breast, lung, and prostate cancers—ErbB3 has become an important focus in cancer biology and targeted therapy research.
