| In Vitro | OATP1A2 exhibited higher affinity for Tebipenem Pivoxil(TBPM-PI)(K(m)= 41.1 μM)than OATP2B1(K(m)> 1 mM)for this agent. TBPM-PI has high intestinal apical membrane permeability[1]. In mouse，rat，dog and monkey，TBPM-PI were absorbed quickly，and the bioavailability was(71.4，59.1，34.8 and 44.9%，respectively. [2].For these isolates the MIC at which 90% of isolates are inhibited was significantly lower for TBM than for the reference oral antibiotics，as follows: 0.002 microg/ml for PSSP，0.004 mug/ml for gPISP(2x)，0.016 microg/ml for gPISP(isolates with abnormal pbp1a and pbp2x genes and isolates with abnormal pbp2x and pbp2b genes)，and 0.063 microg/ml for gPRSP[3].Against Citrobacter freundii，Escherichia coli，Klebsiella pneumoniae，and Enterobacter spp. including extended-spectrum beta-lactamase producers，tebipenem had a potent activity with or without ceftazidime-resistance.[4]Chinchillas inoculated via the transbullar approach with Streptococcus pneumoniae serogroup 6 were included in the efficacy study with TBM-PI. TBM-PI resulted in survival rate of 83%，compared with 0% survival for controls(p<0.01). Quantitative cultures in the middle ear effusions at day 5 of the TBM-PI group yielded 3.5+/-2.4log(10)CFUs/ml.[5] |
