Intended use
This immunoassay kit allows for the in vitro quantitative determination of chicken Thyroxine concentrations in serum, plasma, tissue homogenates, cell culture supernates, and other biological fluids.
The ELISA is based on the competitive binding enzyme immunoassay technique. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Thyroxine, During the reaction, Thyroxine in the sample or standard competes with a fixed amount of biotin-labeled Thyroxine for sites on a pre-coated Monoclonal antibody specific to Thyroxine. Excess conjugate and unbound sample or standard are washed from the plate. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then a TMB substrate solution is added to each well. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm ± 2 nm. The concentration of Thyroxine in the samples is then determined by comparing the O.D. of the samples to the standard curve.

目前市场上常见的TA克隆试剂盒，根据连接方法可以分成两种：方法1：以T4连接酶进行连接的方法，这两种方法转化子比较多，阳性率也不错，唯一的不足就是连接的时间长。因为连接时间越长效率越高，一般快速连接方法需要半个小时到一个小时；达到最佳的效果，要过夜连接。方法2：以拓扑异构酶（TOPO）进行连接的方法，这种方法的优势是快速连接，在5-15分钟就可以将目的基因克隆到载体中，阳性率比较高，这种方法国内也有人模仿。这种方法虽然速度快，但是也有一些缺陷，比如长片段连接效率不如T4连接酶，转化子少
