IPTG 是异乳糖模拟物，能够引起乳糖操纵子的转录过程，因此能够诱导乳糖操纵子下游基因对应蛋白的表达。
In Vitro
pull down（诱导蛋白表达，E. coli BL21 cells）
For pull down, plasmid pET-N-HSP70 was transformed into E. coli BL21 cells. The expression ofrecombinant GST-HSP70 protein was induced by isopropyl-β-D-thiogalactose-pyranoside (IPTG, Solarbio, China II0130). After the cells are destroyed by ultrasound,GST-tag protein purification kit was used to obtain purified GST-HSP70 proteins. His-G3 protein (0.5 mg) and GST-HSP70 (0.5 mg) were incubated at 4˚C for 30 min,then BeyoMag Anti-GST Magnetic Beads was added to the mixture and incubated at 4˚C overnight. The elution sample was analyzed by SDS-PAGE and Western blotting.

The plasmids were transformed into E. coli BL21 (DE3) for expression study. The cells were grown in LB medium with ampicillin (0.1 mg/mL) at 37℃ until OD600 was 0.6–0.8. Next, protein expression was induced with 0.3 mM isopropyl-β-d-thiogalactoside (IPTG) (Beijing Solarbio Science & Technology Co., Ltd), and the cells were grown at 37℃ for further 3 h.
