抗体依赖性细胞介导的细胞毒作用(ADCC)是一种细胞介导的免疫防御机制,免疫系统的效应细胞主动分解膜表面抗原与特异性抗体结合的靶细胞。典型的ADCC涉及通过抗体激活自然杀伤(NK)细胞。NK细胞在其细胞表面上表达Fc受体,主要是cd16或FcγRIIIa(CD16a)。这些Fc受体识别并结合抗体的Fc部分,如IgG,它已结合到病原体感染的靶细胞表面。一旦Fc受体与IgG的Fc区结合,自然杀伤细胞就会释放细胞因子,如IFN-γ和细胞毒性分子,攻击被病原体感染的靶细胞。人类FcγRIIIa在残基158的位置显示出二态性。一个等位基因(V158)编码一个较高的Fc亲和受体变体,在氨基酸残基158处有缬氨酸,另一个等位基因(F158)编码一个较低的Fc亲和受体变体,在氨基酸残基158处有苯丙氨酸。
