Cas9(产脓链球菌CRISPR相关蛋白9)是一种内切核酸酶，当它被sgRNA(单引导RNA)招募到特定的DNA序列时，引入双链断裂进入DNA。这种双链断裂通过非同源性末端接合或同源重组在哺乳动物细胞中修复。非同源性末端接合通常导致切割位点上几个碱基对的缺失或插入，当导致移码时，导致目标基因的功能失活。表达cas9的Daudi细胞可以用靶向感兴趣基因的sgRNA转导或电穿孔以快速产生敲除细胞库或细胞系。
