JAK(Januskinase)/STAT(SignalTransducerandActivatorofTranscription)通路被多种细胞因子和生长因子激活,在细胞生长、造血和免疫反应中起重要作用。在哺乳动物中,有四种JAKs(JAK1,JAK2,jak3和TYK2)和七种STAT蛋白。干扰素α(IFNα)与其受体的结合导致jak1和tyk2的激活,其反过来磷酸化并激活stat1和STAT2。磷酸化的stat1和2形成异二聚体并与IRF9/p48结合,形成称为isgf3的蛋白质复合物。该复合物易位至细胞核并与启动子区域中的ISRE(干扰素刺激的响应元件)结合,从而促进干扰素诱导型基因的转录。
