【产品介绍】
常规细胞培养需要添加血清来维持细胞的增殖能力,尽管血清可以满足一般细胞培养需求,但在进行营养成分优化和标记特定成分进行研究的实验中,仍需要去除某些特定的背景成分,常用的去除方法有碳吸附。
碳吸附胎牛血清用于体外培养,去除背景值以验证激素的作用效果,研究包括类固醇受体连接,细胞受体类固醇调节,各种组织激素分泌和甲状腺激素功能。生产过程中,使用炭和右旋糖苷去除血清中的激素。
血清通常用作细胞生长的添加物,拥有多种功能和用途,如:富含大分子蛋白,低分子量的营养素,作为氨基酸、碳水化合物、维生素的额外来源;提高介质的缓冲能力;可结合或中和有毒成分;减少细胞氧化应激,减少细胞凋亡;提供促进生长的因子,有助细胞适应培养环境。
【适用范围】
碳吸附胎牛血清常用于体外培养研究验证激素的作用效果。
适用于:培养干细胞、原代细胞、神经细胞、人类癌细胞、人类骨髓细胞、白血病细胞、人类胚肺成纤维细胞、人类脑神经细胞和瘤细胞等细胞。
【使用方法】
血清从冰冻区中取出后(切勿直接将血清从-20℃进入37℃解冻,这样因温度改变太大,容易造成蛋白质凝集而出现沉淀),置于2℃~8℃中使之融化,解冻过程中间歇地轻轻摇晃均匀,待全部溶解后再分装或直接使用。完全融解的血清从4℃拿出,按要求比例(5%-20%)加基础培养基配置成完全培养基,建议其置于4℃冰箱存放并于1-2周内用完。

【储存条件及保质期】应贮存在-10℃~-20℃,避免反复冻融;有效期为5年。

【注意事项】
血清沉淀的物质主要有:纤维蛋白(絮状沉淀),脂蛋白,冷凝集素,玻连蛋白,胎球蛋白,磷酸钙(显微镜下小黑点沉淀),胆固醇等。
哪些原因容易导致血清沉淀:
1、温度改变太大,血清由-20℃直接放入37℃解冻,因温度改变剧烈容易导致沉淀。
2、血清在室温或4℃冰箱放置时间太久,血清中的不稳定成分会被破坏,影响细胞生长效果;导致血清品质变差,也会产生沉淀。
3、血清分装冻存时,注意在不产生气泡的情况下摇晃均匀,避免因血清成分浓度改变产生沉淀。
4、血清灭活后容易产生沉淀,如需使用灭活血清,请按56℃,30分钟,轻微摇晃原则操作,灭活温度过高、时间过长、摇晃不均都容易产生沉淀。
5、如有血清沉淀,分装后进行离心处理,建议400g离心5分钟,取上清使用。

【产品介绍】
常规细胞培养需要添加血清来维持细胞的增殖能力,尽管血清可以满足一般细胞培养需求,但在进行营养成分优化和标记特定成分进行研究的实验中,仍需要去除某些特定的背景成分,常用的去除方法有碳吸附。
碳吸附胎牛血清用于体外培养,去除背景值以验证激素的作用效果,研究包括类固醇受体连接,细胞受体类固醇调节,各种组织激素分泌和甲状腺激素功能。生产过程中,使用炭和右旋糖苷去除血清中的激素。
血清通常用作细胞生长的添加物,拥有多种功能和用途,如:富含大分子蛋白,低分子量的营养素,作为氨基酸、碳水化合物、维生素的额外来源;提高介质的缓冲能力;可结合或中和有毒成分;减少细胞氧化应激,减少细胞凋亡;提供促进生长的因子,有助细胞适应培养环境。

适用于:培养干细胞、原代细胞、神经细胞、人类癌细胞、人类骨髓细胞、白血病细胞、人类胚肺成纤维细胞、人类脑神经细胞和瘤细胞等细胞。
【使用方法】
血清从冰冻区中取出后(切勿直接将血清从-20℃进入37℃解冻,这样因温度改变太大,容易造成蛋白质凝集而出现沉淀),置于2℃~8℃中使之融化,解冻过程中间歇地轻轻摇晃均匀,待全部溶解后再分装或直接使用。完全融解的血清从4℃拿出,按要求比例(5%-20%)加基础培养基配置成完全培养基,建议其置于4℃冰箱存放并于1-2周内用完。

【储存条件及保质期】应贮存在-10℃~-20℃,避免反复冻融;有效期为5年。

【注意事项】
血清沉淀的物质主要有:纤维蛋白(絮状沉淀),脂蛋白,冷凝集素,玻连蛋白,胎球蛋白,磷酸钙(显微镜下小黑点沉淀),胆固醇等。
哪些原因容易导致血清沉淀:
1、温度改变太大,血清由-20℃直接放入37℃解冻,因温度改变剧烈容易导致沉淀。
2、血清在室温或4℃冰箱放置时间太久,血清中的不稳定成分会被破坏,影响细胞生长效果;导致血清品质变差,也会产生沉淀。
3、血清分装冻存时,注意在不产生气泡的情况下摇晃均匀,避免因血清成分浓度改变产生沉淀。
4、血清灭活后容易产生沉淀,如需使用灭活血清,请按56℃,30分钟,轻微摇晃原则操作,灭活温度过高、时间过长、摇晃不均都容易产生沉淀。
5、如有血清沉淀,分装后进行离心处理,建议400g离心5分钟,取上清使用。
