    Leukemia inhibitory factor (LIF) is a 20 kD protein belonging to the IL-6 receptor family. It binds to a heterodimer membrane receptor composed of LIF specific subunit gp190 or LIFR and subunit gp130 shared with other members of the IL-6 family. The LIF complex receptor emits signals through STAT3 and JAK kinases. LIF expression was observed in various tissues, including thymus, lung, and neuronal tissue. LIF exhibits various biological effects, but the most well-known one is its ability to inhibit the differentiation of mouse embryonic stem cells and promote stem cell self-renewal. Human and mouse LIFs have 79% sequence homology and exhibit cross species activity. However, the inhibition of stem cell differentiation by LIF seems to be mouse specific. It is involved in inducing hematopoietic differentiation of normal and myeloid leukemia cells, inducing neuronal cell differentiation, regulating mesenchymal to epithelial transformation during renal development, and may also play a role in immune tolerance at the mother infant interface. Selective splicing transcript variants encoding multiple isomers have been observed for this gene. LIF can be stimulated by pro-inflammatory cytokines such as TNF- α And IL-17 were up-regulated, and LIF levels were found to be elevated in patients with rheumatoid arthritis, nerve injury, systemic inflammation and tuberculosis.

    Molecular Mass
    The 180 amino acid recombinant protein has a predicted molecular mass of 19.6 kD. The predicted N-terminal amino acid is Ser.

    >98%, as determined by Coomassie stained SDS-PAGE and HPLC analysis.
    Formulation    Lyophilized, carrier-free.
    Endotoxin Level    Less than 0.1 ng per µg of protein.

    Storage & Handling
    Unopened vial can be stored at -20°C or -70°C. For maximum results, quick spin vial prior to opening. Reconstitute in water to a concentration of 0.1-1.0 mg/ml. Do not vortex. It is recommended to further dilute in a buffer, such as 5% Trehalose, and store working aliquots at -20°C to -80°C. Avoid repeated freeze/thaw cycles.

    ED50 is ≤ 0.1 ng/ml, corresponding to a specific activity of ≥ 1.0 x 107 units/mg as determined by the dose-dependent stimulation of the proliferation of human TF-1 cells.
