For the determination of Human HBsAg concentrations in serum, plasma and other related biological fluids.

| 工作原理 | 本试剂盒采用的是双抗体夹心法酶联免疫吸附检测技术（ELISA）。测定样品中人乙型肝炎病毒表面抗原水平。向预先包被了抗人乙型肝炎病毒表面抗原抗体的酶标孔中，加入标准品和样本，温育后，加入生物素标记的抗乙型肝炎病毒表面抗原抗体。再与HRP标记的链霉亲和素结合，形成免疫复合物，再经过温育和洗涤，去除未结合的酶，然后加入显色底物TMB，产生蓝色，并在酸的作用下转化成最终的黄色。最后，在450 nm处测定反应孔样品吸光度（OD）值，样本中的人乙型肝炎病毒表面抗原浓度与OD值成正比，通过绘制标准曲线计算出样本中人乙型肝炎病毒表面抗原的浓度。 |
| Purpose | This assay employs the Sandwich enzyme immunoassay technique. Determine the level of Human HBsAg in the sample. Add standard and sample to the microplate that have been pre-coated with Anti-Human HBsAg antibody. After incubation, add Biotin-conjugated anti-Human HBsAg antibody. It is then combined with HRP-conjugated streptavidin to form an immune complex, then incubated and washed to remove unbound enzyme, and then added to the chromogenic substrate TMB to produce a blue color, and converted to the final yellow under the action of acid. Finally, the absorbance (OD) value was measured at 450 nm. The concentration of Human HBsAg in the sample was proportional to the OD value. The concentration of Human HBsAg in the sample can be calculated by drawing a standard curve. |

一、ELISA的原理 ELISA的基础是抗原或抗体的固相化及抗原或抗体的酶标记。结合在固相载体表面的抗原或抗体仍保持其免疫学活性，酶标记的抗原或抗体既保留其免疫学活性，又保留酶的活性。在测定时，受检标本（测定其中的抗体或抗原）与固相载体表面的抗原或抗体起反应。用洗涤的方法使固相载体上形成的抗原抗体复合物与液体中的其他物质分开。再加入酶标记的抗原或抗体，也通过反应而结合在固相载体上。此时固相上的酶量与标本中受检物质的量呈一定的比例。加入酶反应的底物后，底物被酶催化成为有色产物，产物的量与标本中
