蛋白表达及纯化：
1.培养500ml哺乳动物细胞，然后将重组质粒转染到细胞中，通过瞬时表达产生目标蛋白。
2.收集含有目标蛋白的部分（细胞或培养上清）。
3.通过亲和，离子交换，疏水及凝胶过滤等多种层析方法纯化表达的重组蛋白。
4.通过SDS-PAGE电泳检测每步结果并控制最终产品质量。
5.通过Western-blot验证目标蛋白正确性。
交付内容：纯化好的目标蛋白及纯化报告。可按客户要求提供含纯化标签（Tag）或切除纯化标签（Tag）的最终蛋白，纯度最高可达90%以上。
操作步骤：
Ⅰ 实体组织蛋白的提取
1． 在每1mL 冷Lysis Buffer加入10 μL磷酸酶抑制剂， 1 μL蛋白酶抑制剂和 5μL 100mM PMSF，混匀。冰上保存数分钟待用。
2． 每100mg固体组织置于培养皿中，手术剪剪碎成3mm×3mm左右的小块，加入0.5～1 mL冷 Lysis Buffer，玻璃匀浆器上下手动匀浆15次，注意低温操作；
3． 取组织匀浆液转移到1.5mL预冷的离心管，离心10000转/分，4℃离心5 min；
4． 取上清转移至新的预冷的离心管中，即为全蛋白提取物，蛋白定量（Bradford法、BCA法）；
5． 分装保存于-70℃,避免反复冻融。
Ⅱ 培养细胞蛋白提取
1． 贴壁培养的细胞，吸去培养基后，加入10mL/150mm培养板的冷PBS洗两次，每次振摇数次以尽量去除培养液；
2． 悬浮培养的细胞或用细胞刮子刮下的贴壁细胞，将细胞及培养液移至离心管中， 1000转/分离心10 min，再用10mL/150mm培养板的冷PBS，1000转/分离心5 min洗两次；
3． 在每 1mL冷 Lysis Buffer加入10μL磷酸酶抑制剂， 1μL蛋白酶抑制剂和5μL 100mM PMSF，混匀。冰上保存数分钟待用。
4． 细胞洗涤后，转至新的预冷的离心管中，加入上述配制好的冷Lysis Buffer，加入量如下表所示：

6． 14,000rpm，4℃离心15min，取上清为全蛋白提取物, 蛋白定量（Bradford法、BCA法）；
