| 描述 | 组织微环境中有复杂的细胞组成,这些细胞的表型、状态、丰度、分布都有着重要的生物学意义和临床价值。借助抗体染色可以将其在组织原位上呈现出来。免疫组化染色是一种研究组织形态和原位蛋白表达的常见技术,常规IHC检测只能展示单一指标,难以呈现复杂的组织微环境中的细胞组成、状态和关系,而这些信息对疾病的诊断和治疗至关重要! 酪氨信号放大技术原理:类似常规免疫组化的DAB显色方法,TSA技术同样采用HRP标记的二抗,HRP催化加入体系的荧光素底物,生产活化荧光底物,活化底物可与抗原上的酪氨酸共价结合,使样品上稳定的共价结合荧光素。之后用热修复法洗去非共价结合的抗体,在换下一种一抗来第二轮孵育,换另一种荧光素底物,如此往复就可实现多重标记。 |
| 检验原理 | 多重荧光免疫组化染色是基于抗原、抗体特异性结合的原理,选用辣根过氧化物酶(HRP)标记的二抗抗体,对试剂盒中的荧光染料进行活化,将信号共价结合到抗原上。借助于不同的荧光染料标记,通过多轮染色循环实现组织或细胞的原位多靶点染色。 |

| 使用方法 | 1.荧光染料的稀释:若染料为干粉,使用100ulDMSO溶解,制成100X染料母液;若染料为液态表示出厂即为DMSO溶解的100X母液;使用信号放大反应液按1:100稀释制备染料工作液(现用现配);DAPI使用无菌水按1:100稀释制备工作液。 2.二抗的使用:该试剂盒搭配为抗兔二抗,实验前请核实一抗种属是否匹配。鼠兔通用二抗请参考(#abs50013)。 |

| 注意事项 | 1. 本试剂盒只用于免疫组化,不做其他用途。 2. 本试剂盒仅限于专业人员使用。 3. 应采用适当的防护措施,以避免试剂同皮肤和眼睛接触。 4. 超过有效期的试剂活性可能降低,因此不得使用超过有效期的试剂。 5. 若将本试剂盒的染色组分与其他公司的产品混合使用,在染色过程中可能出现异常情况。 6. 脱蜡不彻底,容易影响染色效果。 7. 为了防止可能出现的假阴性、假阳性结果,实验过程中需有阳性对照及阴性对照同时进行。 8. 使用本试剂盒中所产生的各种废弃物都应按照《医疗废物管理条例》进行处理。 |
