以离子交换树脂或离子交换纤维素等为固定相的一种层析。广泛用于氨基酸、肽、蛋白质、核苷酸、核酸以及生物体的许多离子性的物质（中性糖作为与硼酸的复合物）的分析。此层析中的样品成分的动态主要是由与固定相中的解离基间的静电相互作用的大小来决定的，但许多也是不能忽视吸附力等其它因素的影响。
