Disposable Bioreactors for Inoculum Production and Protein Expression

Although glass-stirred or stainless-steel-stirred reactors dominate in the area of animal cell cultivation, users are increasingly trying to integrate disposable bioreactors wherever possible. Today applications range from inoculum to glycoprotein-production processes, which are most commonly performed in various membrane bioreactors and the Wave in small- and middle-scale production. For this reason we present protocols instructing the reader how to handle the CeLLine, the miniPerm, and the Wave. In our experience, the methods described for a Chinese hamster ovary (CHO) suspension cell line can also be successfully applied to other animal suspension cells, such as insect cells (Sf-9) and human embryogenic kidney cells (HEK-293 EBNA cells).
