| 检测原理 | The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Human DCP. Standards or samples are added to the appropriate microtiter plate wells then with a biotin-conjugated antibody specific to Human DCP. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Human DCP, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulp huric ac id solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Human DCP in the samples is then determined by comparing the OD of the samples to the standard curve. |

试剂，只需与H2O2溶液混和即成应用液，可直接作底物使用。另外，TMB又有无致癌性等优点，因此在ELISA中应用日趋广泛。酶反应用HCL或H2SO4终止后，TMB产物由蓝色呈黄色，可在比色计中定量，最适吸收波长为405nm。ABTS虽不如OPD和TMB敏感，但空白值极低，也为一些试剂盒所采用。另一种HRP的底物为3-(4-羟基)苯丙酸[3-(4-hydroxy)phenly propionic acid,HPPA]，经HRP作用后，产物显荧光，可用荧光光度计测量。用于ELISA的优点为可加宽定量
