一、 GFP背景概述 绿色荧光蛋白（green fluorescent protein, GFP）最早由Osamu Shimomura于1962年在水母（Aequorea victoria）（图2a）中发现。这种蛋白质在蓝色波长范围的光照激发下发出绿色荧光，其发光过程需要冷光蛋白质Aequorin的帮助，而且，这个冷光蛋白质可与钙离子（Ca2+）相互作用（图3）。在Aequorea victoria中发现的野生型绿色荧光蛋白的分子量较小，仅为27~30kDa，而编码GFP的基因序列也很短
