9N 2.0 DNA连接酶
| 9N 2.0 DNA连接酶（又名9oN DNA Ligase）能够催化磷酸二酯键的形成，使与同一互补靶 DNA 链杂交的两条相邻寡核苷酸链的5´-磷酸末端和3´-羟基末端通过磷酸二酯键相连。该酶来源于极度耐热的海底超嗜热古菌（Thermococcus sp.）9°N 菌株中克隆的连接酶基因，在 45℃-90℃ 范围内均有活性。该酶进一步经基因工程基因突变，使得耐受更高的温度，在90℃孵育15min仍然保留50%以上的活性，因此其适用于：（1）高温条件下，连接DNA链上的切刻位点；（2）与 PCR 反应条件兼容可用连接酶检测反应；（2）连接酶链式反应，对等位基因进行特异性检测。 | ||||||
