本试剂盒可测各种动物血清（浆）、组织、培养细胞、细胞培养上清等样本中碱性磷酸酶活性。碱性磷酸酶分解磷酸苯二钠，产生游离酚和磷酸，酚在碱性溶液中与4-氨基安替吡啉作用经铁化钾氧化生成红色醌衍生物，根据红色深浅可以测定酶活力的高低。
碱性磷酸酶AKP几乎存在于身体各种组织中，是膜结合酶。肠上皮、肾小管、成骨细胞、肝脏、胎盘及白细胞中尤其丰富。血清中AKP主要来源于肝和骨。AKP与肠内脂质转移及骨质钙化有关。血清AKP测定对肝胆系统及骨骼系统疾病的研究有参考意义。
优点如下：
1、快速简便：全程约40分钟，可测80例左右样本；
2、取样量微：常规操作取样量50l， 酶标仪操作仅需5l即可测样本中的AKP活力；
3、稳定性好：试剂盒2～8℃存放3个月有效；
4、再现性好：变异系数CV=1.2%,20倍稀释线性仍然良好；
5、回收试验： X =99%；
6、受外界影响因素小：干扰因素少，重复性强；
7、测试面广：可测动物血清(浆)、组织、各种体液、灌流液、各种培养细胞以及细胞培养上清液等。
注意事项：
1. 本试剂盒的检测试剂中含有萤光素酶，反复冻融会导致其逐渐失活。尽管经测试本试剂反复冻融5次对于其检测效果无显著影响，为取得良好的使用效果，*次解冻后可适当分装保存。反复冻融过程中，可能会导致检测试剂中出现少量沉淀，此时宜平衡至室温，并尽量溶解。如仍有残留的不溶物，可以离心去除后使用，经测试不会影响后续的检测效果。
2. 检测时需使用白色或黑色的96孔板或384孔板。如果使用普通透明的96孔板或384孔板，相邻孔之间会产生相互干扰。
3. 使用说明中提供了检测ATP标准品的方法，实际检测细胞活力时通常并不需要检测ATP标准品。

碱性磷酸酶AKP几乎存在于身体各种组织中，是膜结合酶。肠上皮、肾小管、成骨细胞、肝脏、胎盘及白细胞中尤其丰富。血清中AKP主要来源于肝和骨。AKP与肠内脂质转移及骨质钙化有关。血清AKP测定对肝胆系统及骨骼系统疾病的研究有参考意义。
