线粒体呼吸链复合物I（NADH－辅酶Q 还原酶）活性比色法定量检测试剂是一种旨在使用合成辅酶Q 同功类似物和特异性抑制剂，通过反应系统测定样品中还原型烟酰胺腺嘌呤二核苷酸（NADH）氧化后峰值的降低，即采用比色法测定样品中酶活性的权威而经典的技术方法。该技术经过精心研制、成功实验证明的。其适合于各种纯化线粒体样品（动物、人体、酵母）以及细胞或组织裂解悬液样品的还原型烟酰胺腺嘌呤二核苷酸（NADH）－辅酶Q 还原酶的特异性活性检测。其用于衰老、能量代谢、蛋白组学、病理生理学、神经病变等研究。产品不含污染性蛋白酶，严格无菌，即到即用，操作简捷，性能稳定，反应优化，检测敏感。
线粒体呼吸链复合物I，通常称为还原型烟酰胺腺嘌呤二核苷酸辅酶Q还原酶（reduced nicotinamide adeninedinucleotide coenzyme Q reductase；NADH-CoQ reductase），又称为还原型烟酰胺腺嘌呤二核苷酸脱氢酶（reduced nicotinamide adenine dinucleotide dehydrogenase；NADH dehydrogenase），是线粒体电子传递链中最大的结构成分：含有30至40多个多肽结构。其特征性的酶活性是鱼藤酮敏感的NADH－辅酶Q还原酶（NADH:Q Reductase）。复合物I催化线粒体内电子由供体NADH传递到内膜上辅酶Q受体（泛醌；ubiquinone）的能量转移反应，为整个呼吸链反应系统的第一步。基于辅酶Q底物，在鱼藤酮存在与否的情况下，通过NADH－辅酶Q还原酶的催化，转化成还原型泛醌（CoQH2），同时还原型烟酰胺腺嘌呤二核苷酸（reduced nicotinamide adenine dinucleotide；NADH）转化为氧化型烟酰胺腺嘌呤二核苷酸（nicotinamideadenine dinucleotide；NAD），在分光光度仪下产生吸收峰值的变化（340nm 波长），由此定量测定NADH－辅酶Q还原酶的特异活性。

线粒体呼吸链复合物I试剂盒(NADH-辅酶Q还原酶) 使用注意事项
1． 微量试剂取用前请离心集液。
2． Annexin V-EGFP，Propidium Iodide（PI）避光保存及使用。
3． Propidium Iodide （PI）有毒，操作时请戴手套。
4． 本试剂盒适用于检测活细胞，流式细胞仪检测时，细胞数量不应低于1×105，，不推荐用于检测组织样本。
5． 推荐使用悬浮培养细胞。如果是贴壁细胞，需用不含EDTA的胰酶消化，如消化不当，可能引起假阳性，而用细胞刮子会造成细胞粘连成团，而影响检测。可将胰酶消化后细胞的保存在含2%BSA的PBS中，防止进一步的损伤。
6． 细胞固定后可能导致荧光的淬灭，请不要固定样品。
7． 因检测细胞的类型、凋亡诱导剂种类、使用的检测仪器不同，因而流式检测的荧光补偿也不同，因此建议每次检测均需使用未经凋亡诱导处理的细胞作为对照，进行荧光补偿的调节。
