线粒体呼吸链复合物III（辅酶Q-细胞色素C 还原酶）活性比色法定量检测试剂是一种旨在使用还原性合成辅酶Q 同功类似物和特异性抑制剂，通过反应系统测定样品中细胞色素C 还原后峰值的增高，即采用比色法测定样品中酶活性的权威而经典的技术方法。该技术经过精心研制、成功实验证明的。其适合于各种纯化线粒体样品（动物、人体、酵母）以及细胞或组织裂解悬液样品的辅酶Q-细胞色素C 还原酶的特异性活性检测。可用于衰老、能量代谢、蛋白组学、病理生理学、神经病变等研究。产品不含污染性蛋白酶，严格无菌，即到即用，操作简捷，性能稳定，反应优化，检测敏感。
线粒体呼吸链复合物III，通常称为辅酶Q－细胞色素C还原酶、细胞色素还原酶或bc1复合物（ubiquinolcytochrome c oxidoreductase；cytochrome reductase；bc1 complex），是线粒体电子传递链的中心元素：含有十一个亚单位，包括细胞色素b（Cytochrome b），细胞色素c1（cytochrome c1）和Rieske铁硫蛋白（Rieskeiron-sulfur protein），通过Q循环（Q cycle）进行催化作用。其特征性的酶活性是抗霉素敏感的辅酶Q－细胞色素C还原酶。复合物III催化氧化型细胞色素C被还原为还原型细胞色素C，线粒体内电子由供体还原型泛醌（ubiquinol；UQH2）或还原型辅酶Q（reduced Coenzyme Q；CoQH2）传递到细胞色素C上的能量转移反应，进行呼吸链传递。该酶异常会导致心肌疾病。辅酶Q－细胞色素C还原酶反应系统测定氧化型细胞色素C的浓度变化。基于还原型泛醌或还原型辅酶Q底物，在抗霉素存在与否的情况下，通过辅酶Q-细胞色素C还原酶的催化，转化成泛醌（ubiquinone；UQ）或辅酶Q（Coenzyme Q；CoQ），同时氧化型细胞色素C，转化为还原型细胞色素C，在分光光度仪下产生吸光峰值的变化（550nm 波长），由此定量测定辅酶Q－细胞色素C还原酶的特异活性。

线粒体呼吸链复合物III试剂盒(辅酶Q-细胞色素C还原酶) 使用注意事项
1． 微量试剂取用前请离心集液。
2． Annexin V-EGFP，Propidium Iodide（PI）避光保存及使用。
3． Propidium Iodide （PI）有毒，操作时请戴手套。
4． 本试剂盒适用于检测活细胞，流式细胞仪检测时，细胞数量不应低于1×105，，不推荐用于检测组织样本。
5． 推荐使用悬浮培养细胞。如果是贴壁细胞，需用不含EDTA的胰酶消化，如消化不当，可能引起假阳性，而用细胞刮子会造成细胞粘连成团，而影响检测。可将胰酶消化后细胞的保存在含2%BSA的PBS中，防止进一步的损伤。
6． 细胞固定后可能导致荧光的淬灭，请不要固定样品。
7． 因检测细胞的类型、凋亡诱导剂种类、使用的检测仪器不同，因而流式检测的荧光补偿也不同，因此建议每次检测均需使用未经凋亡诱导处理的细胞作为对照，进行荧光补偿的调节。

线粒体呼吸链复合物III（辅酶Q-细胞色素C 还原酶）活性比色法定量检测试剂是一种旨在使用还原性合成辅酶Q 同功类似物和特异性抑制剂，通过反应系统测定样品中细胞色素C 还原后峰值的增高，即采用比色法测定样品中酶活性的权威而经典的技术方法。该技术经过精心研制、成功实验证明的。其适合于各种纯化线粒体样品（动物、人体、酵母）以及细胞或组织裂解悬液样品的辅酶Q-细胞色素C 还原酶的特异性活性检测。可用于衰老、能量代谢、蛋白组学、病理生理学、神经病变等研究。产品不含污染性蛋白酶，严格无菌，即到即用，操作简捷，性能稳定，反应优化，检测敏感。
线粒体呼吸链复合物III，通常称为辅酶Q－细胞色素C还原酶、细胞色素还原酶或bc1复合物（ubiquinolcytochrome c oxidoreductase；cytochrome reductase；bc1 complex），是线粒体电子传递链的中心元素：含有十一个亚单位，包括细胞色素b（Cytochrome b），细胞色素c1（cytochrome c1）和Rieske铁硫蛋白（Rieskeiron-sulfur protein），通过Q循环（Q cycle）进行催化作用。其特征性的酶活性是抗霉素敏感的辅酶Q－细胞色素C还原酶。复合物III催化氧化型细胞色素C被还原为还原型细胞色素C，线粒体内电子由供体还原型泛醌（ubiquinol；UQH2）或还原型辅酶Q（reduced Coenzyme Q；CoQH2）传递到细胞色素C上的能量转移反应，进行呼吸链传递。该酶异常会导致心肌疾病。辅酶Q－细胞色素C还原酶反应系统测定氧化型细胞色素C的浓度变化。基于还原型泛醌或还原型辅酶Q底物，在抗霉素存在与否的情况下，通过辅酶Q-细胞色素C还原酶的催化，转化成泛醌（ubiquinone；UQ）或辅酶Q（Coenzyme Q；CoQ），同时氧化型细胞色素C，转化为还原型细胞色素C，在分光光度仪下产生吸光峰值的变化（550nm 波长），由此定量测定辅酶Q－细胞色素C还原酶的特异活性。

1． 微量试剂取用前请离心集液。
2． Annexin V-EGFP，Propidium Iodide（PI）避光保存及使用。
3． Propidium Iodide （PI）有毒，操作时请戴手套。
4． 本试剂盒适用于检测活细胞，流式细胞仪检测时，细胞数量不应低于1×105，，不推荐用于检测组织样本。
5． 推荐使用悬浮培养细胞。如果是贴壁细胞，需用不含EDTA的胰酶消化，如消化不当，可能引起假阳性，而用细胞刮子会造成细胞粘连成团，而影响检测。可将胰酶消化后细胞的保存在含2%BSA的PBS中，防止进一步的损伤。
6． 细胞固定后可能导致荧光的淬灭，请不要固定样品。
7． 因检测细胞的类型、凋亡诱导剂种类、使用的检测仪器不同，因而流式检测的荧光补偿也不同，因此建议每次检测均需使用未经凋亡诱导处理的细胞作为对照，进行荧光补偿的调节。

可逆性ATP酶，能在外能驱动下逆浓差转运H 。线粒体内膜呼吸链中有三个酶复合体具有质子泵功能，能将H 由内腔转运到外腔，它们是：细胞色素c氧化酶、辅酶QH2 - 细胞色素c还原酶、NADH- 辅酶Q还原酶。细菌的的质膜上普遍有质子泵，有的伴有呼吸链组分。嗜盐菌膜上的菌紫质（bacteriorhodopsin）受光照驱动，可将H 运入菌体内浓集。
复合物Ⅲ又称CoQH2-细胞色素c 还原酶复合物, 总相对分子质量为250kDa。含1个细胞色素c1、1个细胞色素b(有两个血红素基团)、1个铁硫蛋白,其中细胞色素b由线粒体基因编码。复合物Ⅲ催化电子从辅酶Q向细胞色素c传递,并且每传递一对电子,同时传递4个H 到膜间隙。
可以看出CoQ在复合物Ⅰ与Ⅲ，Ⅱ与Ⅲ之间传递还原当量，Cyt c在复合物Ⅲ与Ⅳ之间传递还原当量。他们又将这四种复合物1：1：1：1的比例混合，加上CoQ和Cyt c重组，基本上恢复了线粒体原有的催化能力。 借助上述 实验方法 ，呼吸链各组分的排列顺序已基本明确，但仍有些不一致的看法，其中以CoQ至细胞色素C这一部分研究得还很不清楚，对于Fe-S和CoQ的定位和数量也有争议。 (二)氧化呼吸链 1.NADH氧化呼吸链 人体
