1、融合型表达重组蛋白，一般胞内可溶性的，拟选用亲和层析进行纯化；
2、测定范围：含氮量0.05%-90%； 分泌型表达的重组蛋白，通常体积大、浓度低，因此应在纯化之前采用沉淀或超滤等方法先进行浓缩处理；
3、包涵体型表达的重组蛋白，应先离心回收包涵体；
4、表达在细胞膜和细胞壁之间的间隙中的蛋白质，应用低浓度的溶菌酶，然后再用渗透压休克法释放重组蛋白。
不同纯化方法之间的平衡：
1、分辨率：由选择的方法和层析介质生成窄峰的能力来完成。当杂质和目的蛋白性质相似是，在纯化的zui后阶段分辨率是选择层析方法的重要考量标准。
2、处理量：指在纯化过程中目标蛋白的上样体积和浓度等。
3、速度：在初纯化中是重要因素，此时杂质如蛋白酶必须尽快去除。
4、回收率：随着纯化的进行渐趋重要，因为纯化产物的价值在增加。
