1、融合型表达重组蛋白，一般胞内可溶性的，拟选用亲和层析进行纯化；
2、测定范围：含氮量0.05%-90%； 分泌型表达的重组蛋白，通常体积大、浓度低，因此应在纯化之前采用沉淀或超滤等方法先进行浓缩处理；
3、包涵体型表达的重组蛋白，应先离心回收包涵体；
4、表达在细胞膜和细胞壁之间的间隙中的蛋白质，应用低浓度的溶菌酶，然后再用渗透压休克法释放重组蛋白。
