| 背景介绍 | 白细胞介素4(IL-4)是约18-20kDa的单体细胞因子。小鼠IL-4前体为140个氨基酸,由20个氨基酸组成信号肽,120个氨基酸组成成熟的肽链。成熟的小鼠IL-4与人和大鼠IL-4的同源性分别为43%和63%。
IL-4可诱导幼稚辅助性T细胞(Th0细胞)分化为Th2细胞。Th2细胞受IL-4活化后,随后正反馈分泌IL-4。分泌IL-4诱导Th0细胞分化的细胞类型尚未得到鉴定,但近期研究表明,嗜碱性粒细胞可能是该效应细胞。IL-4与IL-13密切相关,且具有相似的功能。 IL-4还可促使横纹肌肉瘤发生有丝分裂、去分化和转移。在过敏性哮喘病人中可观察到IL-4和其它Th2细胞因子参与了气道炎症。 |
| 检测原理 | 本试剂盒采用双抗体夹心酶联免疫吸附检测技术。特异性抗小鼠IL-4抗体预包被在高亲和力的酶标板上。酶标板孔中加入标准品和待测样本,经过孵育,样本中存在的IL-4与固相抗体结合。洗涤去除未结合的物质后,加入生物素化的检测抗体孵育。洗涤去除未结合的生物素化的抗体,加入辣根过氧化物酶标记的链霉亲和素 (Streptavidin-HRP)。洗涤后,加入信号增强剂孵育,洗涤去除未结合的物质后,再次加入Streptavidin-HRP。洗涤后,加入显色底物TMB,避光显色。颜色反应的深浅与样本中IL-4的浓度成正比。加入终止液终止反应,在450 nm波长(参考波长570-630 nm)测定吸光度值。 |

IL-4是II型辅助T细胞(Th2细胞)分泌的细胞因子。IL-4是一种多效性细胞因子,在调节T、B淋巴细胞和其他类型细胞的增殖、分化、凋亡,促进以Th2细胞为特征的免疫应答过程中发挥重要作用。IL-4多重生物效应依赖于细胞类型和分化状态,研究表明其在介导过敏性疾病、自身免疫性疾病、感染性疾病、肿瘤等疾病的免疫反应中有多重作用,对肿瘤、自身免疫性疾病和感染性疾病等有治疗作用,并且IL-4对疫苗免疫应答具有调节作用。

Located in external side of plasma membrane and extracellular space. Is expressed in several structures, including brain; colon; hemolymphoid system; liver; and placenta. Used to study Sjogren's syndrome; atopic dermatitis; and type 1 diabetes mellitus. Human ortholog(s) of this gene implicated in several diseases, including asthma (multiple); autoimmune disease (multiple); hepatitis B; hepatitis C; and pancreatic cancer (multiple).
