| 背景介绍 | C-X-C基序趋化因子10(CXCL10),又名γ干扰素诱导蛋白10(IP-10),是一个分子量为8.7kDa的蛋白,属于CXC趋化因子家族。多种细胞类型包括单核细胞、内皮细胞和成纤维细胞在响应IFN-γ时可分泌IP-10。IP-10具有多种作用功能,如趋化单核/巨噬细胞、NK细胞和树突状细胞,促进T细胞粘附到内皮细胞,抗肿瘤活性,抑制骨髓集落形成及抑制血管生成。
IP-10与趋化因子MIG拥有共同受体CXCR3,但是在感染的宿主防御过程中发挥不同的作用。IP-10参与过敏性接触性皮炎反应和其它过敏性疾病,在免疫介导的过程、神经系统炎症和阿尔茨海默病中出现。也有研究报道它可参与调节炎症性肺损伤后的纤维增生。 |
| 检测原理 | 本试剂盒采用双抗体夹心酶联免疫吸附检测技术。特异性抗小鼠IP-10抗体预包被在高亲和力的酶标板上。酶标板孔中加入标准品、待测样本和生物素化的检测抗体,经过孵育,样本中存在的IP-10与固相抗体和检测抗体结合。洗涤去除未结合的物质后,加入辣根过氧化物酶标记的链霉亲和素(Streptavidin-HRP)。洗涤后,加入显色底物TMB,避光显色。颜色反应的深浅与样本中IP-10的浓度成正比。加入终止液终止反应,在450 nm波长(参考波长570-630 nm)测定吸光度值。 |
