蛋白构建：A DNA sequence encoding the extracellular domain (Met1-Ser297) of human FCGRT (NP_004098.1) was fused with a C-terminal polyhistidine tagged AVI tag at the C-terminus, constructed the plasmid 1; A DNA sequence encoding the human B2M (NP_004039.1) (Met1-Met119) constructed the plasmid 2. The two plasmids were co-expressed and the human FCGRT/B2M heterodimer was purified. The purified heterodimer was biotinylated in vivo by the Biotin-Protein ligase (BirA enzyme) which is co-expressed.
