Note: P.Aeruginosa, formerly known as Pseudomonas aeruginosa, is a non fermenting Gram-negative bacterium. It is one of the most common bacteria in soil. The thalli are slender and different in length, sometimes in ball rod or line shape, arranged in pairs or short chains. There is a single flagellum at one end of the bacteria. It can be seen that the bacteria are active under the dark field microscope or phase contrast microscope. Pseudomonas aeruginosa is an obligate aerobic bacterium. The growth temperature range is 25 ~ 42 鈩?/span>, and the optimum growth temperature is 25 ~ 30 鈩?/span>. In particular, the characteristic that the bacterium can grow at 42 鈩?/span> instead of 4 鈩?/span> can be identified. It can survive on ordinary medium and produce water-soluble pigments, such as pyocynin and fluorescent water-soluble fluorescein. There will be a transparent hemolytic ring on the blood plate. The bacterium contains O antigen (bacterial antigen) and H antigen (flagella antigen). O antigen contains two components: one is its outer membrane protein, which is a protective antigen; The other is lipopolysaccharide, which has specificity.
Operation:
1. Thaw on ice, then operate in the super clean table.
2. Dip a small amount of glycerol bacteria with sterilized inoculation ring and streak four areas on the solid plate.
3. Put the solid plate upside down in the incubator for overnight.
4. Select a single colony of appropriate size and expand the culture overnight
