GS115 is a Pichia pastoral strain, which belongs to eukaryotic cells. According to the general antibiotics such as prokaryotes and yeast to ampicillin kanamycin is invalid, so in order to prevent Escherichia coli and other prokaryotes strains pollution on yeast, often in the cultivation of adding some ampicillin and kanamycin antibiotic medium to inhibit bacterial growth and contamination. The phenotype of Pichia Pichia GS115 is Mut+, but the GS115 transformant can produce both Mut+ strain and Muts strain. The expression level of target protein in these two transformation lines may be different and unpredictable, so only by experiment can we get the best yeast expression plan.
The suitable growth temperature of Pichia pastoris is 28 to 30 degrees, and the temperature exceeding 32 degrees is harmful to the expression of protein and may lead to cell death. GS115 Pichia pastoris is histidine deficient type (His4 genotype). If histidine gene is carried on the expression vector, it can compensate histidine deficiency of host bacteria, so it can be screened on transformants without histidine. Pichia pastoris GS115 can grow in YPD medium or grow in minimal media supplemented with histidine, but it can not grow in a single minimal media medium.
