    Comments: The cells express 3-hydroxy-3-methylglutaryl-CoA reductase and hepatic triglyceride lipase activities.
    The cells demonstrate decreased expression of apoA-I mRNA and increased expression of catalase mRNA in response to gramoxone (oxidative stress).
    There is no evidence of a Hepatitis B virus genome in this cell line.
    Propagation: ATCC complete growth medium: The base medium for this cell line is ATCC-formulated Eagle's Minimum Essential Medium, Catalog No. 30-2003. To make the complete growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 10%.
    Temperature: 37.0°C
    Atmosphere: air, 95%; carbon dioxide (CO2), 5%
    Subculturing: Protocol:
    1. Remove and discard culture medium.
    2. Briefly rinse the cell layer with 0.25% (w/v) Trypsin- 0.53 mM EDTA solution to remove all traces of serum that contains trypsin inhibitor.
    3. Add 2.0 to 3.0 ml of Trypsin-EDTA solution to flask and observe cells under an inverted microscope until cell layer is dispersed (usually within 5 to 15 minutes).
      Note: To avoid clumping do not agitate the cells by hitting or shaking the flask while waiting for the cells to detach. Cells that are difficult to detach may be placed at 37�C to facilitate dispersal.
    4. Add 6.0 to 8.0 ml of complete growth medium and aspirate cells by gently pipetting.
    5. Add appropriate aliquots of the cell suspension to new culture vessels.
    6. Incubate cultures at 37�C.

        Subcultivation Ratio: A subcultivation ratio of 1:4 to 1:6 is recommended

      又称分裂准备期、 DNA合成后期或第二间期。指细胞周期的间期中的一个时期,即从 DNA合成期( S期)到分裂期( M期)之间的时期。 A. Howard等于 1953年将从 M期到 S期之间的时期定名为 G1 期,从 S期到 M期之间的时期定名为 G2 期。  

    • 如何用流式区分G2期和M期?

      有丝分裂细胞周期各时相的DNA 含量不同,通常正常细胞的G1/ G0 期具有二倍体细胞的DNA 含量(2N) ,而G2/ M 期具有四倍体细胞的DNA 含量(4N) ,而S 期的DNA含量介于二倍体和四倍体之间。因此,用DNA染料处理细胞后,可以将细胞周期各时相区分为G1/ G0 期,S 期和G2/ M 期,并可通过计算获得各时相的百分率。

      cpuyao 请教大家一个问题,我在药物处理后测定细胞的周期,随着药物浓度的增加,G2/M期比例增加(明显,浓度依赖),S期几乎没有变化,而G1期比例减少。是不是说明细胞阻滞在G2/M期?但是G1期和S期的现象怎么解释呢?随后我们测了凋亡,证明药物可以促进细胞凋亡,这些需要怎么联系到一起呢,第一次做,有好多不明白的地方,希望研究周期和凋亡的战友们帮忙分析一下,有没有哪些相关的文献?谢谢大家~ xiaohuilang 细胞周期被阻滞
